New York, NY – Spheryx announces a poster presentation entitled “Detecting and Identifying Free Fatty Acid Breakdown Products of Surfactants Using Total Holographic Characterization” delivered by Dr. Laura Philips, Spheryx President and CEO, on November 15 at the 2022 PEGS Europe Conference. PEGS Europe is a 3-day event that draws international participants interested in collaborating and sharing best practices in the areas of protein and antibody engineering, oncology, expression, analysis, immunogenicity, and therapeutics.

The poster attached below demonstrates the ability of Spheryx’s xSight to identify contaminants in protein solutions. Beyond particle size, xSight measures the refractive index and the shape of particle contaminants to identify their composition. The poster presents measurements of free fatty acids compared to breakdown products of surfactants, and distinguishes these breakdown products from other contaminants such as silicone oil and protein aggregates. These results demonstrate xSight’s ability to detect early signs of surfactant aging, which may limit the shelf-life of biologic pharmaceuticals. The co-authors of the poster are Ms. Juliana Lumer, Ms. Tiffany Markus and Dr. Fook Chiong Cheong, all of Spheryx.

Spheryx will demonstrate xSight, their particle characterization instrument, and xStream, their automated sample-loader, at the annual AAPS PharmSci 360 conference at the Boston Convention & Exhibition Center October 16-19.

October 13, 2022 - New York, NY – Spheryx will continuously demonstrate xSight and xStream in Booth 858 at the Exhibition of the American Association of Pharmaceutical Scientists 2022 PharmSci 360 Conference. The conference will take place from October 16-19, 2022 at the Boston Convention & Exhibition Center in Boston, MA.

xSight is Spheryx’s particle characterization instrument that embodies Spheryx’s proprietary technology, Total Holographic Characterization. xStream is the companion robotic system that automates sample delivery and measurement, enabling total walk-away capability for xSight implementation.

The demonstration will show xSight’s unique capability to distinguish, identify and count multiple different contaminants in biologic pharmaceutical formulations simultaneously, even when they are the same size. Spheryx publishes a paper in “Scientific Reports” demonstrating Spheryx’s xSight’s ability to measure yeast cell viability and changes in cell structure upon cell death.

July 27, 2022 - New York, NY – Spheryx announces the publication of an article entitled “Label-free Viability Assay using In-line Holographic Video Microscopy” in the peer-reviewed journal, Scientific Reports. The article can be found here. The paper was authored by Rostislav Boltyanskiy, Mary Ann Odete, Fook C. Cheong, and Laura A. Philips, all of Spheryx.

Spheryx’s xSight was used to determine the viability of yeast cells under different stress conditions without the need for fluorescence labels. The cell index of refraction changes upon cell death, generating a characteristic change in the measurement using Spheryx’s proprietary Total Holographic Characterization® technology as embodied by xSight. Changes in the structure of the cell is also observed using xSight.

Spheryx and NIST publish a paper on stressed NIST monoclonal antibody (mAb) protein samples using Spheryx’s xSight and other technologies to evaluate protein aggregate density in the peer-reviewed Journal of Pharmaceutical Sciences.

Spheryx’s xSight was used to determine the density of the monoclonal antibody (mAb) aggregates and the results were found to be similar to measurements made with a fluorescent microscope. xSight is Spheryx’s particle characterization instrument which uses Spheryx’s proprietary Total Holographic Characterization® technology to analyze colloidal mixtures to determine particle size, shape, composition and concentration of multiple particle types simultaneously. New Software Release for Spheryx’s xSight Adds Many New Features Requested by Customers

Spheryx’s new software release adds capabilities to the user interface expanding data analysis capabilities and data reporting formats.

New York, NY – Spheryx announces a new software release for xSight, its particle characterization instrument. xSight is already known for its unique ability to evaluate complex heterogeneous mixtures of particles with a user-friendly interface. The new software release includes multiple new features and incorporates customer feedback and continuous improvement.

Users can now load multiple datasets simultaneously, reducing the time and effort to review results. Multiple datasets can be plotted simultaneously to compare subtle variations between measurements. Users can also customize the automated reports to view specific regions of size and refractive index. Concentrations of multiple components of a heterogeneous mixture can be viewed as cumulative totals, which is particularly valuable for the pharmaceutical industry as per FDA requirements.

Additionally, the user interface for xStream, the robotic automated sample loader for xSight, has been streamlined to ease the transfer of sample information, further enhancing measurement of samples in 96-well plates. Also included in the new software release are enhanced noise reduction, added robustness and a variety of bug fixes.

New Software Release for Spheryx’s xSight Increases Particle Characterization Measurement Speed by 20%

Spheryx’s new software release reduces the time it takes for xSight to characterize particles by 20% as measured from the time the user pushes “start” until the analysis is complete.

New York, NY – Spheryx announces a new software release for xSight, its particle characterization instrument. xSight is known for its unique ability to evaluate complex heterogeneous mixtures of particles using the particle index of refraction. The new software release reduces measurement time by 20% on a standard sample of silicone oil and protein aggregates at concentrations of 105 particles/mL. Particle measurement time is measured from the time the user presses start until the analysis is complete. This standard sample has a wide range of particle sizes (500 nm to 10 µm), and different particle types (spherical and irregularly shaped particles), and is in the mid-range of concentrations (104 – 106 particles/mL) measured by Spheryx’s xSight. The improvement is dependent on the type of particles in the sample and the concentration of particles. Samples with higher concentrations can see a larger reduction in measurement time, some as much as 50%. Also included in the new software release is enhanced noise reduction which is particularly important for sensitive detection of small particles. Improvements in speed and sensitivity will enhance efficiency for applications in biologic pharmaceuticals and semiconductor slurries, as well as other industrial and research applications.

New York, NY – Spheryx, Inc. announced today that the National Center for Advancing Translational Sciences (NCATS) of the National Institutes of Health (NIH) has awarded the company a Phase IIB Small Business Innovation Research (SBIR) grant for $2,000,000 over 2 years. This new grant is a continuation of an earlier grant entitled “Instrument for Characterization of Protein Aggregates” and will support further development of Spheryx’s proprietary Total Holographic Characterization® technology. These highly competitive grants are intended to stimulate technological innovation and commercialization for small businesses.

New York, NY – Spheryx, Inc. announced today that the National Center for Advancing Translational Science (NCATS) of the National Institutes of Health (NIH) has awarded the company a Phase II Small Business Innovation Research (SBIR) grant for $1,500,000 over 2 years to support the modification of their proprietary Total Holographic Characterization® technology to meet the specific needs of the biologics pharmaceutical industry. These highly competitive grants are intended to stimulate technological innovation and commercialization for small businesses.

Spheryx, Inc. was awarded a Supplemental Small Business Innovation Research grant by the National Science Foundation to continue the development of a novel approach to analyzing the physical properties of colloidal dispersions for application to CMP slurries for the semiconductor industry.

New York, NY – Spheryx, Inc. announced today that the National Science Foundation (NSF) has awarded the company a Supplemental Small Business Innovation Research (SBIR) grant. The focus of the funded work is on the application of Total Holographic Characterization™ to CMP slurries used in the manufacturing of semiconductor wafers. This grant is intended to bridge the gap between Phase I and possible Phase II funding. It helps accelerate the Phase I project to the commercialization stage by matching 50% of third party investments, extending the R&D effort beyond the current grant to meet technical requirements of third party investors. The patented technology has the potential to improve quality assurance for CMP slurries and reduce waste in the manufacturing process of semiconductor wafers.

Spheryx Awarded an NSF SBIR Grant to Develop Total Holographic Characterization™ for Commercialization

Spheryx, Inc. was awarded a Small Business Innovation Research grant by the National Science Foundation to support the development of a novel approach to analyzing the physical properties of colloidal dispersions which has diverse applications across multiple industries in the $5 billion particle characterization market.

New York, NY – Spheryx, Inc. announced today that the National Science Foundation (NSF) has awarded the company a Small Business Innovation Research (SBIR) grant. This award will fund the development of a novel approach to analyzing the physical properties of colloidal dispersions, Total Holographic Characterization™, based on holographic video microscopy.

This patented technology, exclusively licensed to Spheryx, will have immediate applications in industries as diverse as pharmaceuticals, cosmetics, personal care products, petrochemicals, coatings and food, all of which rely on the properties of colloidal dispersions. The worldwide market for particle characterization exceeded $5 billion per year in 2012. The funded work extends the state­-of-the-­art in particle characterization by providing simultaneous information about both the size and the composition of individual particles in suspensions. In a matter of minutes, Spheryx’s fast precise holographic measurements build up a clear picture of a sample’s composition, even for complex multicomponent mixtures and blends.

Dr. Laura Philips, President and CEO stated, “This SBIR award is an acknowledgement that these innovations to Total Holographic Characterization, if successful, can be a powerful tool for industry. Access to new dimensions of information will be useful for product development, process control and quality assurance in all industrial sectors that rely on properties of colloidal materials, thereby increasing opportunities for innovation, enhancing product performance, and decreasing manufacturing costs.” Dr. David Ruffner, Director of Analytical Services and Research Scientist added “These funds will be committed to transforming holographic characterization from ideal laboratory samples to real world industrial materials. The improvements will allow us to characterize particles in these non-ideal suspensions with nanometer resolution in size and part per thousand in refractive index.”

No other particle characterization technique offers such a wealth of particle ­resolved information. The delivery of a 100 ­fold acceleration in performance, funded by this grant, will enable applications in large volume and high throughput applications. Spheryx plans to use the resulting real­time capabilities to improve manufacturing efficiency by identifying and helping to correct process deviations and failures. In so doing it will reduce product costs in all of the industrial sectors that develop and sell colloidal materials.

Spheryx will give poster presentations at PittCon, the world's largest annual conference and exposition for laboratory science, on March 11, 2015 at 1 pm in New Orleans, and at the American Chemical Society National Meeting and Exposition on March 22nd, 2015 at 6 pm in Denver.

New York, NY – Spheryx, Inc. will present at two upcoming meetings, the PittCon 2015 Conference and Expo, the world’s largest annual premier conference and exposition on laboratory science, and the 249th American Chemical Society National Meeting and Exposition. The presentations will cover Spheryx’s technology for Total Holographic Characterization™ based on holographic video microscopy developed in the laboratory of Prof. David Grier, Professor of Physics at New York University, Director of NYU's Center for Soft Matter Research, and co-founder of Spheryx. The authors of the presentation are Spheryx President, CEO and co-founder, Dr. Laura Philips, Dr. David Grier and Dr. Fook Cheong, also a co-founder of Spheryx. Dr. Philips will be making the presentations.

The presentation at PittCon will take place at 1 pm on March 11, in Hall F of the Ernest N. Morial Convention Center, New Orleans, LA. Spheryx has completed a successful seed round financing, enabling the company to move into new space, and to advance the commercial development of Total Holographic Characterization™. Ruffner, PhD, an internationally noted expert in holographic video microscopy with deep ties to the Grier research group at NYU, to be Director of Analytical Services and Research Scientist.

The new funds have already been put to work as Spheryx expands with a move into new space and the hiring of David B. Spheryx Awarded Phase IIB NIH SBIR Grant to Support the Development of Total Holographic Characterization® for Biologic Pharmaceutical Applications

Spheryx, Inc. was awarded a Phase IIB Small Business Innovation Research grant by the National Institutes of Health to support the development of their Total Holographic Characterization® for applications in the pharmaceutical industry, particularly protein-based pharmaceuticals.

New York, NY – Spheryx, Inc. announced today that the National Center for Advancing Translational Sciences (NCATS) of the National Institutes of Health (NIH) has awarded the company a Phase IIB Small Business Innovation Research (SBIR) grant for $2,000,000 over 2 years. This new grant is a continuation of an earlier grant entitled “Instrument for Characterization of Protein Aggregates” and will support further development of Spheryx’s proprietary Total Holographic Characterization® technology. These highly competitive grants are intended to stimulate technological innovation and commercialization for small businesses.

Previous funding from Phases I and II of this SBIR grant resulted in two publications in the Journal of Pharmaceutical Sciences (here, here), demonstrating the capabilities of Spheryx’s technology to monitor contaminants in biologic products, with Spheryx’s first particle characterization product, xSight.
